镉致大鼠肝细胞凋亡及N-乙酰半胱氨酸的保护作用 被引量:7

Cadmium induced apoptosis in rat hepatocytes and cytoprotective effect of N-acetyl-cysteine

摘要 用两步灌流法获得大鼠肝细胞,应用四甲基偶氮噻唑蓝(MTT)比色法、Hoechst33258荧光染色法和流式细胞仪检测醋酸镉对肝细胞存活率和凋亡率的影响,以及N-乙酰半胱氨酸(NAC)的保护效应。结果表明,肝细胞暴露于浓度为2.5、5、10μmol/L的醋酸镉后,细胞相对存活率显著下降(P<0.01),凋亡率显著升高(P<0.05),且具有剂量-效应关系;2mmol/L的NAC可使细胞相对存活率明显提高(P<0.01),凋亡率下降。结果提示,一定浓度醋酸镉可引起肝细胞凋亡,NAC具有一定的保护作用。 Rat hepatocytes were isolated by a two-step perfusion technique.Applying MTT assay,Hoechst 33258 and flow cytometry detected hepatocytes cell viability and apoptosis rate induced by cadmium acetate.And the effects of N-acetyl-cysteine(NAC) on cell viability and apoptosis rate were investigated.The results showed that cell viability were decreased(P0.01)and apoptosis rate were increased(P0.05)in a dose-dependent manner after exposure to 2.5,5 and 10 μmol/L cadmium acetate.2 mmol/L NAC increased cell viability(P0.01)and decreased apoptosis rate.It suggests that cadmium acetate may induce the apoptosis of hepatocytes at certain doses and NAC has cytoprotective effect on cadmium.
