摘要 以一株隐球酵母为材料,考查了酸热法中盐酸浓度、盐酸用量、酸热时间、无水乙醇用量、有机溶剂单因素对油脂提取收率的影响,并通过正交试验确定了0.5g干菌体用10mL 4mol/L盐酸沸水浴处理8min,加入无水乙醇用量10mL,用乙醚12mL和石油醚12mL萃取时,油脂得率最高;并对提取的油脂进行了气相色谱分析,结果表明:棕榈酸为20.78%,硬脂酸为4.57%,油酸为59.9%,亚油酸为11.36%,亚麻酸为0.23%,其他3.16%。 The acid-heating extraction was a simple and efficient method of lipid extraction. The single factor such as hydrochloric acid concentration, hydrochloric acid dosage, processing time by hydrochloric acid, alcohol dosage and organic solvent were determined, and the best extraction condi- tions through orthogonal experiment were selected for further study. The results were as follows: aether 10 ml and petroleum ether 10 ml, hydrochlo- ric acid concentration 4 mol/L, hydrochloric acid dosage 10 ml, processing time by hydrochloric acid 8 rain, alcohol dosage 12 ml. It's content fatty acid were palmitic acid 20,78 %, stearic acid 4.57 %, oleic acid 59.9 %, linoleic acid 11.36, linolenic acid 0.23 %, others 3.16 %.
