己酮可可碱对大鼠脑缺血再灌注损伤细胞凋亡及Bcl-2、Caspase-3表达的影响 被引量:2

摘要 目的:探讨己酮可可碱对大鼠局灶性脑缺血再灌注损伤Bcl-2、Caspase-3表达的影响。方法:SD大鼠随机分为假手术组、脑缺血-再灌注模型组、川芎嗪组和己酮可可碱组,每组10只。术前实验性灌胃给药,川芎嗪给药剂量为50 mg/kg,PTX组给药剂量为100 mg/kg,每日一次,连续给药10天。采用线栓法制作大鼠脑缺血-再灌注损伤模型。造模成功2 h后再尾静脉注射1次。假手术组和对照组给予生理盐水。用TUNEL法检测脑缺血2 h再灌注24 h大鼠皮质和海马神经细胞凋亡率,免疫组化法观察Bcl-2和Caspase-3蛋白表达。结果:与脑缺血-再灌注组比较,川芎嗪组和PTX组皮质和海马部神经细胞凋亡率明显减少(P均<0.01),Bcl-2表达明显增加(P均<0.01),Caspase-3表达明显降低(P均<0.01),川芎嗪组和PTX组两组间比较差异无统计学意义(P>0.05)。结论:PTX可抑制大鼠神经细胞凋亡,促进Bcl-2表达,降低Caspase-3表达。 Objective : To investigate the effect of pentoxifylline on Bcl-2, Caspase-3 expression on focal cerebral ischemia-reperfusion in- jury rats. Methods : SD rats were randomly divided into four groups : sham operation group, cerebral ischemia - reperfusion model group and TMP group and the pentoxifylline group. 10 in each group. The rats with drugs treatment were undertaken middle cerebral artery oc- clusion(MCAO) and reperfution after administrating drugs consecutively for 10 days via intragastric administration and administrated a- gain by tail vein injection at 2h after operation. TMP administered a dose of 50mg/kg, PTX group administered a dose of 100mg/kg. Sham-operated group and the control group received normal saline. Apoptosis rate in rat cortex and hippocampus neuronal were observed By TUNEL assay,and Bcl-2 and Caspase-3 protein expression were detected by immunohistoehemical. Results:Compared with cerebral ischemia-reperfusion group, the rates of nerve cells apoptosis in the TMP group and the PTX group were significantly reduced ( P all 〈 0.01 ), Bcl-2 expression were significantly increased ( P all 〈 0.01 ). Caspase - 3 expression were significantly lower ( P all 〈 0.01 ). Between the two groups of TMP and PTX, there was no statistically difference ( P 〉 0.05 ). Conclusion : PTX could inhibit neuronal cells apoptosis in rats,promote Bcl-2 protein expression and reduce Caspase-3 protein expression.
