摘要
目的利用Morris水迷宫和自主活动仪检测6-羟基多巴(6-hydroxydopamine,6-OHDA)致帕金森病(Parkinson's disease,PD)大鼠空间学习记忆能力和精神行为的改变。方法淘汰有记忆障碍的大鼠及有旋转行为的大鼠,应用立体定向技术单侧黑质致密部以及中脑腹侧被盖区注射6-OHDA,3周后腹腔注射阿朴吗啡(apomorphine,APO)0.5 mg/kg诱导旋转,旋转圈数>7 r/min的大鼠归入PD模型组,旋转圈数≤7 r/min的大鼠为未成模组。最后共纳入实验大鼠74只,模型组40只,未成模组19只,正常对照组15只。应用Morris水迷宫测定大鼠的寻台潜伏期、游泳速度、游泳距离及穿越平台次数;应用自主活动测评其精神行为。免疫组织化学观察黑质酪氨酸羟化酶阳性多巴胺能神经元变化。结果 PD模型组大鼠与未成模组和正常对照组相比,寻台潜伏期显著增加(P<0.01),未成模组和正常对照组相比差异无统计学意义(P>0.05)。游泳速度明显下降,游泳距离显著增加(P<0.05)。PD模型组穿越平台次数比未成模组(P<0.05)和正常对照组(P<0.01)显著减少。模型组大鼠的自主活动次数明显少于正常对照组(P<0.01)。PD组大鼠黑质TH表达明显减少。结论单侧黑质致密部及腹侧被盖区注射6-OHDA所致PD大鼠不仅有APO诱导的旋转行为,而且存在学习记忆能力的下降,并伴有精神行为障碍。
Objective To observe the learning and memory ability and mental behavior in 6- hydroxydopamine (6-OHDA)-induced Parkinson's disease model rats by Morris water maze test and autonomic activities test. Methods Rats without memory impairment and circle behavior were injected into compact of substantia nigra and mesencephal ventral tegmental area with 6-OHDA unilaterally by stereotactic technique, and then were induced by apomorphine (APO)with grou were test. p. The latency to find the platform, swimming speed, swimming distance and crossing times detected with Morris water maze. The mental behavior was detected by autonomic activities Immunohistochemistry was used to observe the change of tyrosine hydroxylaseimmunoreactive(TH-IR) positive neurons in substantia nigra. Results Compared with failedmodel group and normal group,the escape latency of rats in the PD model group was significantly increased ( P〈0.05), but there was no significant difference between failed-model group and normal group( P 〉0.05). The swimming speed was decreased and the swimming distance was increased. The crossing times of rats in PD model were significantly decreased compared with failed-model group ( P 〈0.05) and normal group( P 〈0.01). The autonomic activity frequency in PD model was significant less than normal group ( P 〈0. 001). The expression of TH-IR cells in substantia nigra of PD model group was obviously decreased. Conclusion The PD model by injection of 6-OHDA into compact of substantia nigra and mesencephal ventral tegmental area unilaterally not only has behavioral impairment, but also deficits in learning and memory activity and in mental behavior.
