南方红豆杉组织培养及紫杉醇的产生 被引量:32

摘要 南方红豆杉茎段愈伤组织在不同基本培养基及激素组合的培养基上,其平均鲜重生物量具显著差异,而干重生物量无差异。适宜浓度的NAA可以取代常用的、但具致癌活性的2,4-D。TA或B5基本培养基附加1.0mg/LNAA和0.5mg/LBA为其愈伤组织生长的适宜配方。经有机溶媒提取、硅胶柱层析分离、薄层层析检识证明愈伤组织中含有紫杉醇和10-脱乙酰基巴卡丁-Ⅲ。 Fresh weight biomass of callus originated from Taxus chinensis var. mairei young stem explant on different media were significantly different according to Duncans multiple range test,but the dry weight biomass of calli had no difference.The proper concentration of NAA could be a suitable alternative of 2,4 D,a commonly used plant growth regulator but had risk of inducing cancer.The superior media were TA or B 5 supplemented with 1.0 mg/L NAA and 0.5 mg/L BA.TLC results of organical extractions of calli and concentrations of Si gel column chromatographic fractions indicated that taxol and 10 deacetylbaccatin Ⅲ were existed.
