摘要
南方红豆杉茎段愈伤组织在不同基本培养基及激素组合的培养基上,其平均鲜重生物量具显著差异,而干重生物量无差异。适宜浓度的NAA可以取代常用的、但具致癌活性的2,4-D。TA或B5基本培养基附加1.0mg/LNAA和0.5mg/LBA为其愈伤组织生长的适宜配方。经有机溶媒提取、硅胶柱层析分离、薄层层析检识证明愈伤组织中含有紫杉醇和10-脱乙酰基巴卡丁-Ⅲ。
Fresh weight biomass of callus originated from Taxus chinensis var. mairei young stem explant on different media were significantly different according to Duncans multiple range test,but the dry weight biomass of calli had no difference.The proper concentration of NAA could be a suitable alternative of 2,4 D,a commonly used plant growth regulator but had risk of inducing cancer.The superior media were TA or B 5 supplemented with 1.0 mg/L NAA and 0.5 mg/L BA.TLC results of organical extractions of calli and concentrations of Si gel column chromatographic fractions indicated that taxol and 10 deacetylbaccatin Ⅲ were existed.
