摘要
目的通过星点设计法优化红豆杉细胞悬浮培养中的同步化条件。方法以磷酸盐阻断、放线菌酮和低温 3种同步化方法为考察因素 ,通过对细胞分裂指数测定结果进行二项式拟合描绘三维效应面 ,从效应面上选取较佳的实验条件。结果二项式拟合数学模型复相关系数较高 ,R2 =0 .97,最佳同步化方法为磷酸盐阻断11.5 74d ,放线菌酮 0 .2 90mg/ml,温度 8.379℃。结论星点设计结合效应面法可用于优化红豆杉细胞培养的同步化条件 ,且效果良好。
PurposeRotational central composite design was employed to optimize the overall desirability of synchronization of Taxus chinensis cells.MethodsIndependent variables of the experimental design were three synchronization method, double phosphate starvation, cycloheximide and low temperature. Dependent variable was the index of cell division (Y). Overall desirability was fitted to a second-order polynomial equation, through which three dimensional response surface graphs were produced. Optimum experimental conditions were selected from the stationary point of the response surfaces.ResultsThe regression coefficient of the nonlinear estimation was higher than 0.97. The optimum synchronization conditions of Taxus chinensis cells were: double phosphate starvation 11.574 days,cycloheximide 0.290 mg/ml and temperature 8.379℃.ConclusionCentral composite design combined with response surface methodology can be used to represent the most desirable effect of synchronization in suspension cell culture of Taxus chinensis.
