孔雀绿比色法同步测定大鼠成骨细胞膜Ca^(2+)-ATP酶和Na^+、K^+-ATP酶活性 被引量:8

摘要 为探索同步测定大鼠成骨细胞膜Ca2+-ATP酶和Na+、K+-ATP酶活性的方法。采用孔雀绿比色法测定磷以反映酶活性。结果显示:Ca2+浓度以及成骨细胞传代培养的代数是影响酶活性的重要因素。Ca2+有助于提高ATPase活性。Ca2+浓度为40μmol/L时,ATPase活性较高。细胞传代越多,膜Ca2+-ATPase活性越低。成骨细胞膜Ca2+-ATPase活性低于Na+、K+-ATPase。结论:孔雀绿比色法同步测定大鼠成骨细胞膜Ca2+-ATP酶和Na+、K+-ATP酶活性,方法简便、灵敏、可靠。 The aim of this study was to find a method to measure simultaneously the activities of Ca 2+ ATPase and Na +, K + ATPase of rat osteoblast membrane. In this test, Ca 2+ ATPase and Na +, K + ATPase released inorganic phosphate (Pi) from ATP, and Pi was determined by colorimetric method using malachite green dye to show ATPase activities. Ca 2+ concentration and osteoblast culture generation were important factors affecting ATPase activities. Ca 2+ could elevate membrane ATPase activity. When Ca 2+ concentration reached 40 μmol/L, ATPase activities were improved. Ca 2+ ATPase activity decreased with the increase of osteoblast culture generation. Ca 2+ ATPase activity was lower than Na +, K + ATPase activity in osteoblast membrane. This method is simple, sensitive and reliable.
