摘要
目的动态检测角膜碱烧伤后血清抗角膜变性蛋白抗体的水平,了解其变化特征。方法新西兰白兔13只,制作单眼角膜中央部中度碱烧伤模型,制备正常及碱烧伤角膜蛋白提取液,以酶标法(ELISA)检测角膜碱烧伤前后不同时期血清抗角膜变性蛋白抗体水平。结果角膜碱烧伤后机体产生特异性IgG抗体,2周升高明显,5~6周达高峰,之后下降,至8周时仍然明显高于烧伤前水平。再次烧伤角膜,抗体产生更为强烈。结论体液免疫参与了角膜碱烧伤后的病理过程。
ObjectiveTo detect and monitor serum levels of the antibodies against denatured corneal proteins following corneal alkali burns.MethodsOf 13 male New Zealand rabbits,9 received a moderate alkali burn with a filter paper soaked in 1mol/L NaOH to the centre of the right cornea.8 wk later,the left cornea received the same burn.The sera were collected weekly.In the same way,the sera of 4 normal rabbits were collected.The normal or burned corneas were excised and homogenized in PBS.The supernatant was collected after the homogenate was centrifuged.The corneal supernatant and sera were subjected to enzyme linked immunosorbent assay(ELISA) to examine the level of the antibodies against denatured corneal proteins.ResultsThe denatured corneal proteins stimulated the body to create antibodies which began to increase at 2 wk following corneal alkali burns,peaked in the 5th or 6th wk,and then dropped.A second corneal alkali burn on the other cornea during the 8th wk might cause a faster and more intenser response than that found after a single burn.ConclusionCorneal alkali burns stimulate a response of the humoral immune system which creats the specific antibodies against denatured corneal proteins.The antibodies in serum follow a regular course:appearing,rising,peaking,dropping,and then rising again faster and more intense following a second corneal alkali burn.
