摘要
目的研究缺氧对视网膜色素上皮(retinalpigmentepithelium,RPE)细胞的影响。方法采用细胞计数及免疫组织化学的方法,将RPE细胞接种于24孔培养板,分别放入正常和缺氧环境中,37℃进行培养。不同时间后进行计数及抗FOS蛋白抗体染色。结果缺氧组的细胞计数均高于正常组(P<0.05),抗FOS蛋白抗体染色,缺氧组明显强于正常组。结论缺氧可以明显促进RPE细胞的增殖,其机理是通过诱导c-fos癌基因的表达而起作用。
ObjectiveTo investigate the effect of hypoxia on the growth of bovine retinal pigment epithelial(RPE) cells.MethodsThe RPE cells were passaged into 24well dishes,and the dishes were put into normal or hypoxic chamber,respectively.After different times,the cells were counted.At the same time,antiFOS protein antibody was used to study the cfos oncogene expression.ResultsOf the hypoxia group,the cell numbers were more than those of normal group at all time points(P<0.05),and the staining of antiFOS protein antibody was stronger than normal.ConclusionHypoxia can stimulate the growth of RPE cells.It may act through inducing cfos oncogene expression.
