缺氧对牛视网膜色素上皮细胞增殖的促进作用 被引量:3

Stimulative Effects of Hypoxia on Bovine Retinal Pigment Epithelial Cell Proliferation and cfos Oncogene Expression in vitro

摘要 目的研究缺氧对视网膜色素上皮(retinalpigmentepithelium,RPE)细胞的影响。方法采用细胞计数及免疫组织化学的方法,将RPE细胞接种于24孔培养板,分别放入正常和缺氧环境中,37℃进行培养。不同时间后进行计数及抗FOS蛋白抗体染色。结果缺氧组的细胞计数均高于正常组(P<0.05),抗FOS蛋白抗体染色,缺氧组明显强于正常组。结论缺氧可以明显促进RPE细胞的增殖,其机理是通过诱导c-fos癌基因的表达而起作用。 ObjectiveTo investigate the effect of hypoxia on the growth of bovine retinal pigment epithelial(RPE) cells.MethodsThe RPE cells were passaged into 24well dishes,and the dishes were put into normal or hypoxic chamber,respectively.After different times,the cells were counted.At the same time,antiFOS protein antibody was used to study the cfos oncogene expression.ResultsOf the hypoxia group,the cell numbers were more than those of normal group at all time points(P<0.05),and the staining of antiFOS protein antibody was stronger than normal.ConclusionHypoxia can stimulate the growth of RPE cells.It may act through inducing cfos oncogene expression.
