软坚消瘿汤对甲状腺细胞凋亡及Bcl-2/Bax表达的影响 被引量:2

摘要 目的观察软坚消瘿汤对实验性自身免疫性甲状腺炎(EAT)大鼠甲状腺凋亡蛋白Bcl-2/Bax和细胞凋亡的影响,从细胞凋亡角度探讨其作用机制。方法运用甲状腺球蛋白与弗氏佐剂混合免疫注射法,联合饮用高碘水模拟大鼠实验性自身免疫性甲状腺炎。免疫组化染色法检测大鼠甲状腺细胞Bcl-2/Bax变化,原位末端标记技术检测细胞凋亡。结果软坚消瘿汤能显著降低Bax表达,增加甲状腺细胞Bcl-2表达,减少甲状腺细胞凋亡。结论软坚消瘿汤可能通过降低滤泡上皮细胞Bax表达,提高Bcl-2表达从而减轻甲状腺细胞凋亡。 Objective:To evaluate the therapeutic effect of Xiaoying Ruanjian Decoction on experimental autoimmune thyroiditis (EAT) from the levels of apoptosis and apoptosis-regulated proteins Bcl-2/Bax of EAT rats. To Investigate the action Mechanism from apoptosis. Methods: An animal model of experimental autoimmune thyroiditis (EAT) rats ~bas developed by injecting thyroglobulin mixed with Freund's adjuvant immunization and drinking Periodate water. To observe the level change of Bcl-2/Bax in the thyroid of rat with Immunohistoehemieal staining. To detect the levels of apoptosis with TUNEL. Results: Ruanjian Xiaoying Decoction significantly reduces the levels of the expressions of Bax and apoptosis in thyroid ceils and increases the expression of Bcl-2. Conclusion: Ruanjian Xiaoying Decoction alleviates the levels of Bax in thyroid cells and increas the expression of Bcl-2 by the suppression of relative expressions of antibodies in serum levels of EAT, which reduces apoptosis in thyroid cells.
