摘要
为改进大肠杆菌血清型鉴定方法，用５株标准菌株在９６孔Ｕ型板上进行微量凝集试验，优选了反应及观察条件：每孔中加５０μＬ菌体抗原（浓度为１．５×１０９－２．０×１０９个菌／ｍＬ）和５０μＬＯ抗原单因子血清（稀释度从１／１０－１／８０）混合，于３７℃反应１．５－２ｈ，阳性结果可在黑色背景上清晰观察到片状凝集。试验各株均与原血清型一致。用此法对１７株来自山东某地的鸡大肠杆菌进行了血清型鉴定，结果Ｏ７８为５株，Ｏ３６５株，Ｏ１３１４株，Ｏ７０３株，与以前用常规方法对该地区菌型鉴定结果相符。
Five reference strains were used in agglutination test using 96 U well microplate for the improvement of E.coli serotyping.Reaction and observation conditions were optimized:Each well was added with 50uL somatic antigen(1.5×10 9～2.0×10 9 cells/mL)and 50μL(single factor serum of O antigen(dilution ranged between 1/10 and 1/80)and incubated at 37℃for 1.5～2 hours after mixing. Scale like agglutination was observed under dark background if there were positive results. 17 E.coli isolates from a poultry farm in Shandong Provine were serotyped using the method :5 were indentified as O78,5 as O36,4 as O131 and 3 as O70,The results were in conformity with those previously identified in the region.
