摘要
目的:探讨青藤碱、环孢素及两者联合应用对大鼠肝移植模型排斥反应的影响。方法:正常组:8只Wistar,为空白对照。建立SD-WistarOLT模型,分为4组,每组8只SD-Wistar OLT大鼠。对照组、CsA组、SIN组、亚剂量CsA+SIN组。检测血浆IL-2、IL-10含量,肝内TGF-β1表达,肾脏TGF-β1mRNA表达,肝脏内淋巴细胞的凋亡,肝脏的病理损害。结果:SIN+CsA组抑制IL-2及促进TGF-β表达弱于CsA组,促进IL-10分泌强于CsA组,同时促进了肝内淋巴细胞的凋亡,减轻了肝脏的病理损害。结论:SIN能够减轻肝脏移植后的肝脏损害;与CsA联合作用减少CsA的用量,防止肾纤维化的发生。
Objective: To compare the effect of CsA, SIN and their combination on the rejection reaction after the orthotropic liver transplantation in rats. Methods: The blank group was eight Wister rats. We established the stable orthotropic liver transplantation ( OLT ) model. The rat models were randomly divided into four groups and every group was eight SD-Wister rats, Control group, CsA treated group, SIN treated group, SIN and half-value dose CsA treated group. Enzyme-linked immunosorbent assays was carried out to detect the level of IL-2, IL-10. The expression of TGF-β protein in liver and TGF-β mRNA in kidney used immunohistochemisty staining and RT-PCR, respectivly. The lymphocyte apoptotic index in header district of liver was detected using the TUNEL.The liver pathological lesion was scored with Williams's system after HIM staining. Results: The level of IL-2 and expression of TGF- β 1 in the SIN+CsA-treated group was lower than CsA-treated group, but the level of IL-10 was stronger than the CsA. The lymphocyte apoptotic index in header district of SIN+CsA-treated group was the most obvious and the damage of the transplantation liver was the lightness than other groups.Conclusion: SIN could alleviate the pathological damage after OLT in rats. SIN and CsA combination could reduce the dosage of CsA, and Prevented of renal fibrosis.
