摘要
【目的】研究京尼平苷对大鼠前列腺成纤维细胞增殖的影响,并探讨其机制。【方法】培养SD大鼠前列腺成纤维细胞,取对数生长期的细胞随机分成4组:对照组、京尼平苷10mg/L、20mg/L和40mg/L组。不同浓度的京尼平苷孵育大鼠前列腺成纤维细胞48h后,采用MTT法检测细胞增殖,流式细胞仪检测细胞凋亡,ELISA法测定Ⅰ型胶原、Ⅲ型胶原与转化生长因子β1(TGF-β1)蛋白的含量。【结果】与对照组比较,京尼平苷20mg/L和40mg/L可显著抑制大鼠前列腺成纤维细胞的增殖(P<0.01),促进其凋亡(P<0.01),同时减少TGF-β1的分泌和胶原的合成(P<0.05或P<0.01)。【结论】京尼平苷可抑制大鼠前列腺成纤维细胞的增殖,可能与其减少TGF-β1的分泌和胶原的合成、促进前列腺成纤维细胞的凋亡有关。
[Objective] To investigate effects and mechanism of geniposide on proliferation in fibroblast of rats.[Methods]The fibroblast of rats were isolated.The fibroblast which were in exponential phase of growth were divided randomly into 4 groups:control group,geniposide 10mg/L,20mg/L and 40mg/L groups.After incubated with different dosages of geniposide,the proliferation of fibroblast was detected by thiazole blue(MTT)colorimetric assay.Apoptosis was analyzed by flow cytometry.The content of collagen Ⅰ,Ⅲ and TGF-β1 were measured by the ELISA.[Results] Compared with control group,the proliferation of prostate fibroblast in rats were inhibited significantly by geniposide 20mg/L and 40mg/L respectively(P0.01).Apoptosis was increased(P0.01,respectively)and the content of collagen Ⅰ,Ⅲ and TGF-β1 were decreased(P0.05 or P0.01,respectively)by geniposide 20mg/L and 40mg/L,respectively.[Conclusions] Geniposide can inhibit proliferation of prostate fibroblast in rats.It may be associated with reduced secretion of TGF-β1 and collagen synthesis,promoting prostate fibroblasts apoptosis.
