旋覆代赭汤对大鼠胃平滑肌细胞收缩及信号传导通路的影响 被引量:11

Effects of Xuanfu Daizhe Decoction on contraction induced and the pathways of signal transduction in Gastric Smooth Muscle Cells in Rats

摘要 [目的]观察不同浓度的旋覆代赭汤含药血清对大鼠胃窦平滑肌细胞(SMC)收缩及钙离子(Ca2+)浓度的影响,探讨其对胃SMC收缩及信号传导的作用机制。[方法]分离大鼠单个胃窦SMC细胞,分别加入不同浓度含药血清(5%、10%、20%)和三磷酸肌醇(IP3)受体阻滞剂(肝素),镜下观察含药血清对胃窦SMC收缩作用,检测细胞Ca2+总浓度。[结果]旋覆代赭汤含药血清刺激胃窦SMC,随剂量的增加细胞收缩活动逐渐增强(P<0.05),肝素可抑制旋覆代赭汤含药血清对胃窦SMC的收缩作用(P<0.05),各剂量组与无钙缓冲液组的细胞内Ca2+总浓度相比差异无统计学意义。[结论]旋覆代赭汤含药血清可使胃窦SMC收缩,其机制可能是通过IP3信号途径来介导细胞内质网释放Ca2+,使细胞内Ca2+升高,引起胃窦SMC收缩。 [Objective]To observe the effect of different concentrations of serum from rats medicated with Xuanfu Dai zhe Decoction on cell contraction and intracellular calcium(Ca^2+) concentration of gastric antral smooth muscle cells(SMCs) in rats.[Methods]Research on effects of the different concentrations of serum from rats medicated with XuanFu Daizhe Decoction on the contraction of antral SMCs,and influences of inosamine triphosphate receptor inhibitor IP3(heparin)and intracellular Ca^2+ by culturing single antral SMCs.[Results]Serum from rats medicated with XuanFu Daizhe Decoction could significantly stimulate the contraction of gastric antral SMCs in a concentration-dependent manner(P0.05).Heparin could inhibit these contractile action.After cell contraction was induced,the total intracellular calcium concentration showed no significant difference among each serum treatment group and control group.[Conclusion]Serum from rats medicated with XuanFu Daizhe Decoction can obviously induce antral SMC contraction in rats.The mechanism is related to increased calciumion through IP3 calcium ionchannel receptor in the sarcoplasm.
