摘要 获得高质量DNA是进行分子生物学研究的基础。通过CTAB法提取瓜叶菊叶、茎、花的基因组DNA,提取的DNA质量良好,电泳条带清晰,提取过程无明显的DNA降解,分子量接近21Kb。以提取的DNA为模板,用一对引物扩增瓜叶菊中与花色相关的特异基因,得到条带单一、大小与已知一致,说明提取的DNA可以满足相关的分子生物学研究。CTAB法提取瓜叶菊不同组织的DNA产率不同,其中叶的DNA产率最大,茎的产量居中,花的最低。 Obtaining of high quality DNA was the premise of the research of molecular biology. By means of CTAB method, genomic DNA with better quality was extracted from leaf, stem and flower of Senencio, and the electrophoresis strips were clear, and there was no DNA decomposition during the extractions, MW was close to 21 Kb. Taking the extracted DNA as template and using a pair of primers to amplify the specific genes related with colors of flower of Senencio cruentus, DNA with single strips and MW consistent with known was got, which showed that the extracted DNA was good enough for the research of molecular biology. The DNA outputs extracted by CrAB method from different tissues were different, and the output was highest for leaf, intermediate for stem and lowest for flower.
