摘要
用操作性条件反射的方法观察D-半乳糖亚急性中毒大鼠学习记忆行为的影响。将Wistar大鼠进行1w的操作性条件反射训练,达到稳定操作状态后,将其分为对照组和实验组,实验组再分为3个半乳糖剂量组,分别用D-半乳糖12mg,24mg,和48mg·kg-1·d-1,sc,共6w。各组均为6只大鼠。用药期间,每天进行了30min操作行为FR4的测试。记录操作潜伏期,操作比率及30min的辨别能力测试。结果显示,用药3w后,各剂量组大鼠的操作比率明显下降,潜伏期明显延长(P<0.01),辨别性操作能力逐渐下降。上述改变以48mg·kg-1剂量组最为显著。这提示,D-半乳糖亚急性中毒状态下能干扰大鼠的学习记忆的巩固、再现和辨别能力,这种学习记忆的损害与文献报道的D-半乳糖诱导的拟衰老变化是一致的。
The learning and memory impairment were measured by using microcomouter-controlled operant apparatus in aging rat model induced by D-galactose.D-galactose after being administered for six weeks,the mimetic aging model was established and it's effects on fixed ration schedule-controlled behavior were observed.It was found that the latent period of operation was remarkably prolonged(32±10 sec)comparing with controlled ones (2±1 sec),and the behavior of pressing the lever was disrupted and the rate of the response was reduced. Discrimination was also observed in the experiment and the D-galactostreated rats showed a lower ahility to discriminate.The significance of establishing aging-like rat model with memory and learning deficit was discussed.
