摘要
目的:观察苦碟子注射液对高糖损伤后人脑微血管内皮细胞(HBMEC)核因子-κB(NF-κB)、细胞间黏附分子-1(ICAM-1)及血管细胞黏附分子1(VCAM-1)表达的影响,探讨中风病"毒损脑络"病机假说的现代生物学内涵。方法:以培养HBMEC作为靶细胞,用无水葡萄糖制备细胞损伤模型。采用MTT法检细胞的活性;采用免疫细胞化学法和图像定量分析系统观察NF-κB、ICAM-1与VCAM-1蛋白表达的变化。结果:高糖作用24、72h,与同一时间正常对照组比较,高糖组可明显增加NF-κB蛋白在人脑脏微血管内皮细胞的表达(P<0.01)。苦碟子注射液能降低NF-κB、ICAM-1与VCAM-1蛋白的表达,其中10、5、2.5%浓度组保护作用显著(P<0.01)。结论:NF-κB的激活可能为高糖损伤内皮细胞的始动因素之一,其动态变化及与黏附分子相关的毒性效应体现了内毒形成及作用过程。苦碟子注射液能够减少核转录因子与黏附分子的表达,对人脑微血管细胞具有保护作用,为依据"毒损脑络"病机假说所提治法在临床应用提供了生物学佐证。
Objective:To explore the modern biology connotation of the theory of toxin damaging brain collaterals based on the expressive changes of nuclear factor-kappa B(NF-κB), intercellular cell adhesion molecule-1(ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) of human brain microvascular endothelial ceils ( HBMEC ) in high glucose after using of Kudiezi Injection. Methods: Cultured HBMEC was used as target cells, anhydrous glucose was added to the cell medium to prepare the injury model of human endothelial cell. Methyl thiazolyl tetrazolium ( MMT ) method was used to examine the cell activity; lmmunocytochemistry method and image quantitative analysis system were used to detect the expression of NF- κB, ICAM-1 and VCAM-1 of HBMEC. Results:The expressions of NF-κB of HBMEC were enhanced by high glucose after pretreating HBMEC for 24, 72h (P 〈0.01, compared with the normal grounp).Obviously, Kudiezi Injection can decrease the expressions of NF-κB p65, ICAM-1 and VCAM-1, and here, effections of 10%, 5% and 2.5% concentration groups were marked (P〈0.01). Conclusions: Activation of NF-κB p65 was likely to be one of the initiating agents of endothelial cells damnified by high glucose, its dynamic change and correlative toxicity effect of adhesion molecules embodied formation and actions of endogenetic toxins. Kudiezi Injection can decrease the expression of nuclear transcription factors and adhesion molecules and have protection effect to HBMEC, and it provided biology evidence for treating apoplexy with clearing away toxic material and promoting blood circulation methods in clinic.
