摘要
以极大节旋藻基因组DNA为材料,比较了节旋藻RAPD-PCR常用反应体系与2×Taq Master Mix反应体系的不同之处及扩增效果,并对2×Taq Master Mix反应体系进行了优化实验,初步建立了较理想的极大节旋藻基因组DNA的RAPD-PCR反应体系:2×Taq Master Mix 4μL,随机引物1μL,模板DNA 1μL,ddH2O4μL.优化的反应体系操作简便快捷,扩增结果稳定.
The effects were compared between the usual reaction system and 2×Taq Master Mix system of RAPD-PCR for the genomic DNA of Arthrospira maxima.The optimal 2×Taq Master Mix system of RAPD-PCR was established by 2×Taq Master Mix 4μL,template DNA 1μL,Random primer 1μL,and ddH2O 4μL for total volume of 10μL.The optimization system was convenient,rapid and stable.
