肿瘤抗原触发M-CSF和IFN-γ联合基因转染的巨噬细胞抗肿瘤转移作用的研究

摘要 目的本研究探讨了肿瘤抗原触发的M-CSF和IFN-γ基因联合转染的巨噬细胞对黑色素瘤实验性肺转移的治疗作用及其免疫机理。方法将联合基因转染的巨噬细胞经尾静脉回输治疗黑色素瘤实验性肺转移小鼠,经治疗后于荷瘤后第17天处死小鼠,计数荷瘤小鼠的肺部转移结节数并观察其长期存活期,用51Cr4小时释放法检测体外诱导的CTL活性,采用未配对计量资料的t检验处理数据。结果联合基因转染后治疗组的小鼠长期存活率为16%,加用肿瘤抗原触发后的治疗组有33%的小鼠长期存活。小鼠脾细胞经体外诱导的CTL对野生型B16.F10细胞的杀伤活性,在经肿瘤抗原触发的M-CSF和IFN-γ联合基因转染巨噬细胞治疗的荷瘤小鼠最高,与用未经抗原触发的联合基因转染巨噬细胞治疗组差异有显著性(P<0.05)。结论体外经肿瘤抗原触发的M-CSF、IFN-γ联合基因转染的巨噬细胞过继回输疗法是肿瘤特别是转移性肿瘤的一种有效免疫治疗手段,机体的CTL在该疗法的抗肿瘤作用中可能发挥了重要的作用。 Objective To investigate the therapeutic effects and immunological mechanisms of tumor antigen pulsed M CSF and IFN γ gene transfected macrophages on the preestablished tumor bearing mice with pulmonary metastasis. Methods Melanoma bearing mice with experimental metastasis were treated with tumor antigen pulsed M CSF and IFN γ gene transfected macrophages intravenously. Seventeen days later, the pulmonary metastasis nodes were counted and the survival time was observed.The CTL activities were detected by 51 Cr 4 hours release assay,and the student t test was used to evaluated statistical difference. Results Sixteen pecent of the mice treated with M CSF and IFN γ gene cotransfected macrophages survived more than 3 months while 33% of the mice survived after treatment with additional pulse with tumor antigen. After treatment with macrophages pulsed with tumor antigen, pulmonary metastasis nodes of the mice were much less than those of the mice treated with macrophages which were not pulsed with tumor antigen. The CTL activity was the highest in the mice treated with tumor antigen pulsed, gene cotransfected macrophages. It had significant difference when compared with the mice treated with gene cotransfected macrophages without tumor antigen pulse. Conclusions Adoptive infusion of tumor antigen pulsed M CSF and IFN γ gene cotransfected macrophages is an effective method for tumor therapy and the host CTL activity might play the important role in the antimetastatic effect.
