摘要
目的采用高效毛细管电泳法对绞股蓝中的有效成分芦丁和槲皮素同时进行含量测定。方法采用未涂层熔融石英毛细管柱(57 cm×75μm,有效长度50 cm)为分离通道,以12 mmol.L-1硼砂-甲醇(80∶20)为电泳介质,分离电压30 kV,毛细管温度30℃,检测波长360 nm。结果芦丁和槲皮素在20 min内得到很好的分离,分别在0.054~1.070、0.009~0.180 mg.mL-1与峰面积线性关系良好,平均回收率分别为99.0%、98.8%。结论方法简便快速、结果准确、重现性好,可用于绞股蓝及其制剂的质量控制。
Objective To develop a method to determine rutin and quercetin in Gynostemma pentaphyllum by high-performance capillary electrophoresis.Methods An uncoated fused silica capillary(57 cm×75 μm,effective length of 50 cm) was used.The electrolyte buffer was composed of 12 mmol·L-1 borax-methanol(80∶20),voltage 30 kV,the capillary temperature was 30 ℃,and detection wavelength was 360 nm.Results Rutin and quercetin were successfully separated within 20 min,the linear range was 0.054~1.070 and 0.009~0.180 mg·mL-1.The average recovery was 99.0% and 98.8%.Conclusion The method is simple,rapid and reproducible,and could be used for the quality control of Gynostemma pentaphyllum and its description.
