摘要
[目的]研究免疫八肽AT-N的高效液相色谱分析方法。[方法]依据Fmoc固-相多肽合成原理,以不同树脂作为固相载体,分别合成免疫八肽及其乙酰化、酰胺化产物;以水-乙腈为二元梯度洗脱流动相,0.1%三氟乙酸为离子对,用RP-HPLC对合成产物进行纯化,冷冻干燥后得到纯度大于95%的免疫多肽。将这3种多肽混合进行分离度试验。[结果]最终确定了免疫多肽AT-N的HPLC分析方法,使得分离度达到了定量分析要求。[结论]该研究为以后的中试生产提供了理论指导。
[ Objective I The study aimed to research method of highly effective liquid phase analysis of immunity peptide AT-N. [ Method ] According to the principle of Fmoc-solid phase synthesis, respectively took different resins as solid-phase carrier, synthesized immunity peptide as well as its aeetylize, amidation product ; took the water-methyl cyanide as the dual gradient elation flowing and 0.1% trifluoroacetic acid as the ion pair, carried on the purification to the synthetic product with RP-HPLC, the purity of white powdery peptide is bigger than 95%. Fixed them and carried on the experiment of degree of dissociation. [ Result] Determined the HPLC analysis method finally and make the degree of dissociation meet the requirements for quantitative analysis. [ Conclusion ] The method will provide the theory instruction for the later experiment.
