摘要 [目的]建立测定茄子叶中茄尼醇的RP-HPLC分析方法。[方法]采用Agilent 1100 HPLC,Hypersil ODS色谱柱(250mm×4.0mm,5mn),流动相y(甲醇):y(乙醇)为60:40等度洗脱,洗脱流速1.0ml/min,柱温30℃,检测波长210am,保留时间约10.720min。[结果]茄尼醇质量浓度在6.90~220.80μg/m(R=0.9995)范围内线性关系良好,平均加样回收率(n=9)为99.94%,RSD=2.08%。通过与传统提取方法比较,确定最佳的提取方法为微波辐射法。[结论]所建方法具有简便、快速、准确、重复性好的特点,适用于茄子叶中茄尼醇的含量测定。 [Objective] The study aimed to establish a sensitive RP-HPLC method for determining the content of solanesol in eggplant leaves. [Method] The Agilent 1100 HPLC system, Agilent Hypersil ODS column ( A I. D. 250 mm × 4.0 mm, particle size 5 μm) was operated at a column temperature of 30 ℃ .The mobile phase was V (methanol) : V(ethanol) = 60:40 at a flow rate of 1.0 mt/min.210 nm was set as the UV detection wavelength for solanesol, retention time was probable 10.720 min. [ Result ] The results showed a good linear relationship within 6.90- 220.80 pg/ml( R = 0.999 5, the average recovery( n = 9) is 99.94 %, RSD = 2.08 %. Comparison with conventional extraction method, the optimum extraction method was microwave irradiation. [ Conclusion] The method was rapid,simple,accurate and good reproducibility,it was suitable for the determination of solanesol of eggplant leaves.
