垂体腺瘤E-钙粘素和β-连环素的表达及意义

Expression and significance of E-cadherin and β-catenins in pituitary adenoma

摘要 目的:采用免疫组化方法检测人类垂体腺瘤中E-钙粘素和β-连环素的表达,为进一步了解人类垂体腺瘤的生物学行为提供依据。方法:对41例垂体腺瘤病理标本采用免疫组化EnVision法,以单克隆抗体直接检测E-钙粘素、β-连环素的表达。按染色强度和阳性范围评估免疫组化结果,分析粘附分子表达程度与垂体腺瘤的病理亚型、肿瘤侵袭性的相关关系。结果:泌乳素型和生长激素型间,E-钙粘素、β-连环素的阳性表达程度差别均有统计学意义(uc=2.00、2.08,P<0.05);侵袭性、非侵袭性垂体腺瘤间,E-钙粘素、β-连环素的阳性表达程度差别均有统计学意义(uc=2.13、1.98,P<0.05)。结论:E-钙粘素、β-连环素表达与垂体腺瘤的不同病理亚型相关,在生长激素型垂体腺瘤中,存在着E-钙粘素、β-连环素表达明显下调现象;垂体腺瘤侵袭性与E-钙粘素、β-连环素的低表达相关。 Objective:To analyze the expression of E-cadherin and β-catenin in human pituitary adenoma by immunohistochemistry for further understanding of biological benaviors of pituitary adenoma.Methods:The specimens included in this study were collected form 41 pituitary adenoma cases,EnVision was used in immunohistochemical staining,The E-cadherin and β-catenin were direct tested by the monoclonal antibodies.The results were graded depending on the staining intensity and positive range of the immunoreactivity,the correlation between the expression of the E-cadherin and β-catenin with the subtype pituitary adenoma and the invasiveness of pituitary adenoma were analyzed.Results:In the prolactinomas and growth hormone-secreting pituitary adenoma,the positive staining degrees of the E-cadherin,β-catenin were insignificantly different(uc=2.00,2.08 respectively,P0.05;In the invasive and non-invasive pituitary adenoma,the positive staining degrees of the E-cadherin,β-catenin were insignificantly different(uc=2.13,1.98 respectively,P0.05).Conclusion:The expression of E-cadherin and β-catenin in the pituitary adenoma is related to the subtype pituitary adenoma,the expression of E-cadherin and β-catenin in the growth hormone-secreting pituitary adenoma were remarkably decrease.The invasive pituitary adenoma is related to the expression of E-cadherin and β-catenin significantly down-regulated.
