豆类丝核菌中苦马豆素的提取 被引量:21

Abstraction of Swainsonine from Rhizoctonia leguminicola

摘要 采用H2O∶CH2Cl2萃取、732型阳离子树脂交换等方法,从豆类丝核菌中分离到纯结晶,经薄板层析法发现,豆类丝核菌提取物浓缩的粗品液和苦马豆素标准品经Ehrlich's试剂显色后,二者均出现一椭圆形紫色斑点,且Rf值均为0.81;气相色谱法研究发现,提取物纯品标样(15.9420mg/L)与苦马豆素标准品在同一色谱条件下显示出完全一致的峰形,证明从豆类丝核菌中分离到的纯结晶是苦马豆素纯品。研究还表明豆类丝核菌干燥菌丝体中苦马豆素含量达1.23%~2.99%. The crystals of swainsonine were isolated from Rhizoctonia leguminicola by means of extraction between H 2O∶CH 2Cl 2 and a 732 type cation exchang resin for the first time in China.It discoverd that the purple elliptic spots are displayed in thin layer chromatography (TCL) between semifinished product solution of donse abstracts of R.dleguminicola and standardized sample of swainsonine dyed with Ehrlichs reagent and all the volumes of R f are 0.81.The study with gas chromatography (GC) discovers that the same peaks type appear in pure abstract sample (15.942 0 mg/L) and standardized sample of swainsonine under the same conditions,and therefore it is clear that the pure crystals isolated from R.leguminicola is pure swainsonine.The studies show that dry mycelium of Rhizoctonia leguminicola contains 1.23%-2.99% swainsonine,which has opened a new way to obtain swainsonine.
