应用Dot-ELISA进行日本血吸虫循环抗原检测时其滴度与感染度关系的动态观察 被引量:1

摘要 以20条/只和100条/只剂量的尾蚴感染家兔,并于第9wk用吡喹酮对约半数家兔进行一次性灌胃治疗,依此建立日本血吸虫感染模型。用分别定位于日本血吸虫表皮膜、肠道上皮和虫卵的单克隆抗体建立直接斑点酶联免疫吸附试验(Dot-ELISA)并应用该法分别检测感染家兔血中3类循环抗原、膜相关抗原(MAA),肠相关抗原(GAA)和可溶性虫卵抗原(SEA),观察循环抗原滴度水平和日本血吸虫感染度之间的关系。结果发现,当用Dot-ELISA法检测3类循环抗原时,20条尾蚴感染组与100条尾蚴感染组家兔不论治疗与否,血中3类循环坑原滴度水平无显著性差异。 Halves of two groups of rabbits infected separately with two doses of Schistosoma japonicum(each rabbit infected with 20 or 100 cercariae) were treated with praziquentel(220 mg/kg body weight given as a single dose via stomach)at the 9th week after infection. In order to analyse the relationship between titers of circulating antigens(CAg) and infectiosity, we developed the DotELISA based on the application of 3 McAbs to detect respectively the gutassociated antigen(GAA), membrane associated antigen(MAA) and solube egg antigen(SEA) of Schistosoma japonicum. The results showed that the titers of CAgs in sera between the two groups of rabbits had no significant difference, whether they were treated or not. Therefore,DotELISA to detect CAgs is not suitable for judgement of infectiosity of Schistosoma japonicum.
