背景与目的:Twist是一种促进肿瘤转移的转录因子,而KiSS-1为肿瘤转移抑制基因,它们的蛋白表达与非小细胞肺癌(non-small cell lung cancer,NSCLC)的淋巴结转移密切相关。本研究旨在探讨Twist和KiSS-1在NSCLC和正常肺组织中的表达及与患者预后的相关性。方法:收集61例有完整临床病理资料和随访资料的NSCLC标本及15例癌旁正常肺组织标本作为对照,采用免疫组织化学SP法分别检测Twist和KiSS-1的表达,分析其与患者各项临床指标及预后的关系。结果:Twist在NSCLC组织中的表达显著高于癌旁正常肺组织(P<0.05),KiSS-1在NSCLC组织中表达显著低于癌旁正常肺组织(P<0.05);NSCLC组织中Twist及KiSS-1表达均与临床分期、淋巴结转移情况、4年生存率密切相关(P<0.05);Kaplan-Meier生存曲线分析显示患者术后生存率随Twist阳性表达呈现下降趋势,而随KiSS-1阳性表达显著上升。结论:Twist高表达及KiSS-1低表达的患者易于发生淋巴结转移,术后4年生存率较低。
Background and purpose: Twist has been identified as tumor metastasis promoter transcription factor and KISS-1 has been identified as tumor metastasis suppressor gene, and both of them have been identified to be associated with the metastatic potential of non-small cell lung cancer (NSCLC). Our aim was to identify the expression of both Twist and KISS-1 in NSCLC and analyze their correlation with patients' survival. Methods: Immunohistochemical staining using monoclonal Twist and KISS-1 antibody were performed on paraffin embedded specimens from 61 patients diagnosed with NSCLC, and 15 specimens of tumor surrounding lung tissue were used as control. The association with clinicopathologic data and prognosis of NSCLC were analyzed. Results: The expression of Twist was significantly higher in NSCLC than in tumor surrounding lung tissue (P〈0.05), and the expression of KISS-1 was significantly lower in NSCLC than in tumor surrounding lung tissue (P〈0.05). Both of the Twist and KISS-1 expression were positively related with clinical stage, lymph node metastasis and 4 years survival rate of patients postoperatively(P〈0.05). Kaplan-Meier analysis showed that up-regulated expression of Twist was associated with lower survival rate of patients after operation and up-regulated expression of KISS-1 was associated with higher survival rate of patients after operation. Conclusion: Patients with up-regulated expression of Twist and downregulated expression of KISS-1 were prone to have lymph node metastasis and lower 4 years survival rate of patients after operation.
