摘要
目的:研究MMP-9、TIMP-1在化疗前后口腔鳞癌组织中的表达,探讨口腔鳞癌的侵袭机制。方法:应用免疫组织化学SP法检测化疗前后MMP-9和TIMP-1在口腔鳞癌组织中的表达,分析不同临床分期、组织学分级、淋巴结有无转移间的MMP-9及TIMP-1表达是否存在差异,结合临床病理资料和基因蛋白表达特征作差异显著性检验及相关分析。结果:化疗前口腔鳞癌MMP-9表达高于化疗后,MMP-9表达与临床分期,淋巴转移和疗效有关,与组织学分级,性别和年龄无关;化疗前后TIMP-1表达无差异,TIMP-1表达与临床分期,淋巴转移和疗效有关,与分级,性别和年龄无关;MMP-9与TIMP-1阳性率密切负相关(r=-0.54368,P<0.05),化疗前后MMP-9和TIMP-1与疗效之间密切负相关(r=-0.60947,P<0.05)。结论:MMP-9及其抑制因子TIMP-1在口腔鳞癌的进展过程发挥重要作用,综合分析上述基因蛋白表达特征和口腔鳞癌的临床-病理特点可较好地指导治疗。
Objective ： To investigate the expression of MMP-9 and TIMP-1 in oral squamous cell carcinoma （OSCC） before and after intraarterial chemotherapy （IC）, and to explore the invasive behavior, mechanism, and prognosis of OSCC. Methods： Specimens of 20 OSCC patients before and after IC were examined by using SP immunohistochemistry method. The expression characteristics of MMP-9 and TIMP-1, and clinical and pathological features were determined and differences were analyzed. Results： MMP-9 expression in OSCC was higher before IC than after, and significantly related with clinical stages, lymph node metastasis, and curative effect, but not with histological grades, or sex and age of patients; TIMP-1 expression before IC did not differ from that after. There were significant correlations of TIMP-1 expression in OSCC with clinic stages, lymph node metastasis, and curative effect, but not with histological grades, or sex and age of patients ; A significantly negative correlation was observed between MMP-9 positive stainning and TIMP-1 positive staining （ r = - 0. 54368, P 〈 0.05 ）, and there was a closely negative correlation between MMP-9 and TIMP-1 expression with curitive effects of IC （r = -0. 60947, P 〈 0.05 ）. Conclusions： MMP-9 and its inhibitory factor TIMP-1 may play a key role in progress of OSCC. Analysis of the expression characteristics of MMP-9 and TIMP-1 and clinical and pathological features of OSCC may better conduct clinical therapy of OSCC.
