天麻素抗大鼠离体心肌缺血/再灌注损伤的作用 被引量:10

Protective Effects of Gastrodin Against Myocardial Ischemia Reperfusion Injury in Rats

摘要 目的:观察天麻素对离体大鼠心肌缺血/再灌注(I/R)损伤的影响。方法:应用Langendorff离体心脏灌注系统建立心肌缺血/再灌注损伤模型。40只SD大鼠随机分为正常对照组、缺血/再灌注组、天麻素0.1、0.025mmol·L-1组。除正常对照组外,各组分别平衡灌注、全心停灌和再灌注各30min,观察天麻素对左室发展压(LVDP)、冠状动脉流量(CF)、心肌组织超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量以及超微结构的影响。结果:天麻素明显抑制I/R引起的LVDP和CF的下降,显著改善缺血/再灌注后的心功能,且有明显的量效关系。提高心肌SOD活力和降低MDA含量。显著减轻I/R引起的心肌超微结构的损伤性改变。结论:天麻素对I/R引起的心肌损伤有显著的拮抗作用,其机制可能与其增强清除自由基能力,减轻生物膜脂质过氧化反应,维持心肌细胞膜和线粒体的正常功能,改善能量代谢有关。 Objective: To characterize the effect of Gastrodin on the ischemia/reperfusion I/R injury in rat hearts. Methods : The isolated hearts were perfused in the Langendorff mode and were subjected to either continuous perfusion for 90min or 30min for balance, global ischemia and reperfusion respectively. Left ventriculax developed pressure ( LVDP ), coronary flow ( CF ), SOD activity and MDA content in myoeaxdial homogenate, and the myocardial ultrastructure was also observed. Restdts : It was found that Gastrodin cou.Id significantly antagonize the decrease of LVDP and CF induced by UR. Gastrodin could protect endogenous anfioxidant enzymes, such as superoxide dismutase, and inhibit biomembrane lipid peroxidafion, but there was no dose dependent relationship between these two drug groups. It was also found that myocardial ultrastructural changes induced by UR were prevented in Gastrodin group. Conclusion : Gastrodin could protect myocardium against I/R injury in rats ,which might be related with its effects of restoring cardiac function.
