Optimization of the synthesis of N-methyl-β-hydroxyl-phenylethylamine by orthogonal experimental design

摘要 目的优化N-甲基-β-羟基苯乙胺的合成工艺。方法采用正交设计法,重点考察反应原料摩尔比、反应温度、反应时间及后处理时溶剂的量四个因素对收率的影响。结果反应原料摩尔比对反应收率的影响最为显著,其次是后处理溶剂的量和反应时间,影响最小的是反应温度。结论新工艺收率在59.5%-65.5%之间,优化后的工艺条件较温和且操作简便。 Objective To optimize the synthesis of N-methyl-β- hydroxyl-phenyl-ethylamine, which was the key intermediate compound in the synthetic route of clausenamide. Methods Orthogonal experimental design was used to assess the influence of molar ratio of reagent, temperature, reaction time, and solvent on the yield. Results Molar ratio of reagent was more important than reaction temperature. Conclusion: The yield of the improved method was between 59.5% and 65.5%. Up to now, the reaction condition established here was the best way we couht find to prepare the title compound.
