一氧化氮抑制单核-内皮细胞粘附的机制 被引量:5

INHIBITORY MECHANISM OF NITRIC OXIDE ON ADHESION BETWEEN MONOCYTE AND ENDOTHELIAL CELLS

摘要 目的:研究一氧化氮(NO)抑制单核内皮细胞粘附的机制。方法:以硝普钠作为NO的供体,用细胞粘附实验、流式细胞仪荧光强度测定、细胞ELISA、Northernblot等方法分别从细胞、蛋白表达、基因表达方面研究NO对内皮细胞粘附分子(vascularceladhesionmolecule,VCAM1)表达的影响。结果:NO抑制IL1α诱导的单核内皮细胞粘附,抑制IL1α诱发的VCAM1的表达并呈时间依赖性及浓度依赖性,NO在粘附分子VCAM1的基因表达水平上亦呈现抑制作用。结论:NO抑制VCAM1的表达是其抑制单核内皮细胞粘附及抗动脉粥样硬化的机制之一。 Objective: To study the inhibitory mechanism of nitric oxide on adhesion between monocyte and endothelial cells. Methods:Cell adhesion,FACS, cell ELISA and Northern blot were applied to study effects of nitroprusside, a donor of nitric oxide, on expression of protein and mRNA of (vascular cell adhesion molecule, VCAM 1). Results: Expression of VCAM 1 protein and mRNA was inhibited by nitroprusside, in a dose time dependent manner. Conclusion: Inhibitory effect of nitric oxide on VCAM 1 is one of the mechanisms of its antiadhesion between monocyte and vascular endothelial cells,as well as that of its antiatherosclerosis.
