甲醛对大鼠脑组织超氧化物歧化酶和脑细胞DNA损伤影响的研究 被引量:3

摘要 目的研究甲醛染毒对大鼠脑组织超氧化物歧化酶(SOD)和脑细胞DNA损伤的影响。方法采用腹腔注射方式给大鼠染毒,15只大鼠随机均分为对照组(生理盐水)、低剂量甲醛组0.2 mg/kg(1/2000 LD50)、高剂量甲醛组20.0 mg/kg(1/20 LD50);每天1次,染毒7 d。染毒结束后,检测大鼠脑组织SOD的含量,利用单细胞凝胶电泳技术(彗星实验)检测脑细胞DNA损伤。结果高剂量染毒组大鼠脑组织的SOD的活力[(12.89±2.12)U/mg Pro]低于对照组[(16.34±2.68)U/mgPro](P<0.05);腹腔注射甲醛可导致脑细胞DNA片段断裂,高剂量甲醛组彗星尾巴明显长于对照组。结论甲醛可使脑组织SOD活力下降,造成脑细胞DNA的断裂;甲醛对大鼠中枢神经系统具有一定的毒性作用。 Objective To study the effect of formaldehyde on superexide dismutase(SOD) in the cerebral tissues and DNA damage of brain cells of rats. Methods The rats in control group were exposed to 0. 9 % NaCl at 0. 1 ml/10 g, the rats in low and high dose groups were exposed to formaldehyde with 0. 2 and 20. 0 mg/kg, once per day for seven days. Then SOD in the cerebral tissues were tested, and single cell gel electrophoresis technique (comet assay) was used to test the DNA damage of the brain cells. Results The activity of SOD in high dose group [ ( 12. 89 ± 2. 12) U/mg Pro ] showed a significant decrease compared with control group [ ( 16. 34 ± 2. 68) U/mg Pro] (P 〈0. 05). Formaldehyde could significantly cause the break up of the DNA strands. The tail of comet in high dose group was longer than that in control group. Conclusion Formaldehyde can decrease the activity of SOD in cerebral tissues, and cause break up of DNA strands. Formaldehyde has obvious toxic effects on central nervous system in rats.
