摘要
研究了山麦冬嫩叶愈伤组织诱导和分化、不定芽的分化和生根、试管苗移栽和移植所需要的条件,建立起山麦冬嫩叶的无性系技术。结果证明:MS+6-BA0.4 mg.L-1+NAA 1.0 mg.L-1+2,4-D 1.5 mg.L-1是嫩叶愈伤组织诱导的理想培养基;1/2MS+AgNO31.2 mg.L-1+BA0.5 mg.L-1+NAA0.1 mg.L-1是诱导愈伤组织和不定芽分化的理想培养基;1/2MS+IAA0.2 mg.L-1+NAA0.1 mg.L-1是不定芽生根培养理想培养基;移植的试管苗生长旺盛、根系发达,其他生物性状保持不变。
The conditions needed in callus induction,differentiation of axillary bud,cutting and transplantation Liriope spicatalmud been studied,its regenerative clone and rapid propagation line were established. The results showed that the best medium for callus induction was MS+6-BA0.4 mg L^-1+NAA 1.0 mgL^ +2,4-D 1.5 mg L^-1. The best media for differentiation was 1/2MS+AgNO3 1.2 mg·L^-1+ BA0. 5 mg ·L^-1+NAN0. 1 mg·L^-1 , 1/2MS+IAA0. 2 mg· L^-1 +NAA0. 1 mg·L^-1 was suitable for rooting,regeneration. With cinder used as seeding matrixs. Field planting showed that the tube spout grew well with roots flourishing and kept all biological character.
