分子探针在稻瘟病流行病学中的应用研究 被引量:15

摘要 】用重复序列探针MGR586与限制性内切酶EcoR1组合,分别分析了我国1980~1997年在17个省市146块稻田内外的186个不同水稻品种上采集的445个水稻分离菌株及25种不同禾本科植物和杂草上采集的108个非水稻分离菌株的限制性片段长度多态性(RFLPs),依其MGR-DNA指纹的相似率,结合病菌的致病性测定,将表现为48个不同致病型的553个菌株区分为56个谱系。结果表明,稻瘟菌有远距离传播的可能性,并在适宜的气候和人工接种条件下,某些禾本科植物和杂草上的草瘟菌与稻瘟菌可彼此互交。这为稻瘟病的流行和预测提供了新的思路。该文用生物测定和分子生物学相结合的方法,揭示了我国在不同空间和时间的稻瘟菌群体存在着明显的遗传多样性及其流行因素,并对今后的抗病育种及其利用策略进行了讨论。 The MGR-DNA fingerprintings of 475 rice pathogen isolates collected from 186 different rice cultivars and 108 non pathogen isolates collected from 25 different grasses & weeds have been studied with a conversed repetitive sequence probes of MGR 586/EcoR1 digest combination during 1980~1997 in the 146 different rice fields inner & outside. Five hundred and fifty three isolates belonging to 48 pathotypes of Magnaporthe grisea could be distinguished into 56 separate lineages based on MGR-DNA fingerprint similarity. The MGR-DNA fingerprinting and pathotyping indicated that some of the non rice pathogens isolates and rice pathogen isolates could infect each other occasionally under suitable climatic and artificial inoculation conditions, and the fungi had the capability of long distance transmission. The genetic structure and epidemiological tread of M.grisea in one rice growing area can be analysed and the identification method for resistance to rice blast can be improved based on MGR-DNA fingerprint data. The reasonable arrangement and gene rotation of growing rice cultivars with different resistant genes to rice blast will be directed. It puts forward a new idea for forcasting accurately the incidence and epidemic of the rice blast disease.
