地榆鞣质抗肝癌细胞SMMC-7721的MTT及FCM分析 被引量:16

摘要 目的:探讨中药成分地榆鞣质(STM,STL)的抗癌活性,并试图建立一种筛选抗癌药物的方法.方法:运用MTT法测定STM及STL对体外培养人肝癌SMMC-7721细胞的杀伤率井通过流式细胞仪分析细胞分裂周期各时象DNA变化.结果:STM与STL均有明显抗癌活性,与阴性对照组相比有显著差异(P<0.01),并具有剂量效应关系(RM=0.9615,RL=0.9531);STM,STL与MMC联合用药抗癌活性显著增强,与单独用药组相比差异显著(P<0.01);流式细胞仪(FCM)分析人肝癌细胞分裂周期各时象DNA变化显示,药物作用后S期细胞明显减少,增殖指数(PI)降低,并诱导出凋亡峰(AP峰).结论:ST的抗癌作用可能是多方面的,FCM的测定技术为筛选抗癌中药提供新的方法. Aim: To investigate the antitumor effect of Chinese Herb Sanguisorba officinalis tannins (ST) and to find amethod of screening antitumor drugs. Methods: Human liver cancer cell SMMC-7721 cells were cultured in vitro andthen treated with each kind of drug. Tumor cell killing ratewas analyzed by MTT method and DNA content alterationof each phase of cell circle was determined by FCM (flowcytometry)assay. Results: STM and STL both had strong activity of antitumor does^dependent effect (RM =0. 9615, RL= 0. 9531 ). Compared with negative control group, both ofthem had significant difference (P<0.01). Combined withMMC, their antitumor acitivity was stronger than singledrug group (P<0. 01 ). The flow cytometry assay of cell-circle DNA content displayed that S-phase cell DNA contentdecreased significantly. Prolifelation index (PI)delined andapoptosis peak (AP peak)was introduced. Conclusion: FCMprovides a new way of screening the antitumor component ofChinese herb.
