摘要
为了观察地榆鞣质对骨髓抑制的保护作用,本文采用腹腔注射环磷酰胺致小鼠骨髓抑制模型。造模成功后,将骨髓抑制小鼠按体重随机分为:模型组、集落刺激因子组、地榆鞣质20、10、5 mg/kg三个剂量组,另设正常对照组。连续灌胃10 d,每天一次,以外周血液白细胞数量、骨髓细胞DNA含量和CD34+的表达以及骨髓细胞O6-甲基鸟嘌呤-DNA甲基转移酶(MGMT)蛋白的表达为评价指标,研究地榆鞣质对CTX所致骨髓抑制的治疗作用。结果显示地榆鞣质可显著升高小鼠外周血WBC(P<0.05);显著升高小鼠骨髓DNA含量(P<0.05);显著促进CD34+的表达(P<0.05);显著促进MGMT蛋白的表达(P<0.05)。通过本研究可以看出地榆鞣质可显著拮抗CTX所致小鼠骨髓抑制。
The objective of this study was to investigate the protective effect of tannins from Sanguisorba o2Ttcinalis (Great Burner) on myelosuppression in mice, The myelosuppression model was induced by peritoneal injection of cyclo-phosphamide (CTX). Then the myelosuppression mice were randomly divided into:model group, colony-stimulating fac- tor group,three Great Burner tannins treatment groups of different dosage (20, 10,5 mg/kg), and normal group. The drugs were chronically administrated to myelosuppression mice once a day for 21 days. The counts of peripheral blood leukocytes,DNA contents and CD34^+ expression of bone marrow cells and the O^6-methyl guanine'DNA methyltrans-ferase (MGMT) protein expression were used as main indexes and were determined. The results showed that Great Bur- net tannins significantly increased peripheral WBC (P 〈 0.05) ; significantly elevated bone marrow DNA content ( P 〈 0.05 ) ; significantly promoted the expression of CD34^+ ( P 〈 0.05 ) ; also significantly promoted MGMT protein expres-sion (P 〈0.05). Based on these results,it was concluded that Great Burnet tannins can significantly antagonize CTX in- duced myelosuppression in mice.
