摘要
目的:研究健骨固本胶囊对去卵巢大鼠骨质疏松症的防治作用。方法:将SD雌性大鼠行双侧卵巢切除术,随机分为假手术组(Sham组)、模型组(Model组)、GB高剂量组、中剂量组、低剂量组(0.8,0.4,0.2g·kg-1)和阳性对照组仙灵骨葆胶囊(0.8g·kg-1),共6个组,每组10只。12周后,使用Lunar双能X线骨密度测定仪测定全身骨矿量(BMC)和全身骨密度及股骨骨密度;采用市售试剂盒分析血清抗酒石酸酸性磷酸酶(StrACP)含量和骨组织羟脯氨酸含量;股骨三点弯曲实验分析最大应力、弹性模量和能量吸收。结果:健骨固本胶囊可显著增加骨密度、骨矿量、增加股骨干重指数;显著降低血清抗酒石酸酸性磷酸酶水平,提高骨羟脯氨酸含量;明显提高骨最大应力、弹性模量和能量吸收。结论:健骨固本胶囊对大鼠去卵巢实验性骨质疏松症模型具有显著的改善作用。
OBFECTIVE To investigate the effect of Jiangu Guben (JGGB) capsule on postmenopausal osteoporosis in ovari- ectomized female rats. METHODS Sixty female Sprague-Dawley rats without pregnancy and delivery were used, 50 of them were ovariectomized and randomly divided into 5 groups: ovariectomy (OVX), OVX with JGGB capsule 0. 8 g·kg^-1,0. 4 g·kg^-1, and 0. 2 g·kg^-1 , and OVX with Xianlinggubao capsule 0. 5 g·kg^-1, the others 10 rats were sham operated group. The rats were treated on the fifth day after the operation and continuated for 12 weeks. At the end-point of the experiments, the bone mineral density and bone mineral capacity were assayed by double energy X-ray hone-densitometry equipment, the blood was collected to detect the activity of StrACP in serum, and the hydroxyproline contents of femur by commercial kits, the bone's biomechanics of femur were assayed. RESULTS Capsules decreased remarkably the activity of StrACP in serum and increased significantly the contents hydroxyproline of femur, increased significantly the bone mineral density and bone mineral capacity, and improved the bone biomechanical property. CONCLUSION JGGB capsules can effectively prevent the postmenopausal osteopo rosis of ovariectomied rats, the mechanism maybe involved in ameliorated biomechanics and increased the bone mineral capacity and the content of collagen in bone.
