摘要 [目的]研究植酸酶在猪饲料中的应用条件。[方法]采用钒钼酸法测定植酸酶活力,研究植酸酶活力及时间、温度和pH等因素对饲料中植酸酶作用的影响。[结果]各样品溶液的OD平均值分别为0.241、0.245和0.220。总磷量平均值分别为1.6948、1.7229和1.5516μmol/g,酶活平均值分别为5891.78、5743.00和5171.89IU/g。植酸酶的最适温度为55℃。50~65℃下饲料中植酸磷的降解率接近100%,40℃下其降解率为80%。4h后植酸磷的降解率为80%,5~6h后植酸磷基本被完全降解。pH值为2.5和5.5时植酸酶的活性最强;pH值为7.0时,植酸酶的活性很低。在温度为55℃和pH值为5.5的条件下,5h以上植酸酶对植酸钠中植酸磷的降解率为60%-100%;在温度为40℃和pH值为2.5的条件下,4h后其降解率为60%。[结论]该研究为植酸酶在畜禽养殖中的应用提供了理论依据。 [ Objective ] The purpose was to study the application condition of phytase in pig feed. [ Method ] The influences of factors such as phytase activity, time, temperature and pH on the function of phytase in feed were studied by determining the activity of phytase with poly-vanadium-molybdenum acid. [ Result] The average OD values of all the sample solutions were 0. 241,0. 245 and 0. 220 resp. , their average total phosphorous contents were 1. 694 8, 1. 722 9 and 1. 551 6 p, mol/g resp. , their average enzyme activities were 5 891.78, 5 743.00 and 5 171.89 IU/g resp. The optimum temperature for phytase was 55 ℃. The degradation rate of phytate phosphorous in feed was close to 100% at 50 - 65 ℃ and that was 80% at 40 ℃. The degradation rate of phytate phosphorous was 80% after 4 h and the phytate phosphorous was degraded completely basically after 5 - 6 h. When the pH value was 2.5 or 5.5, the phytase was most active ; when the pH value was 7.0, the activity of phytase was very low. The degradation rate of phytate phosphorous in sodium phytate was 60% - 100% while it had been degraded under the condition with temperature and pH value being 55 ℃ and 5.5 for more than 5 h ; its degradation rate was 60% while it had been degraded under the condition with temperature and pH value being 40 ℃ and 2.5 for 4 h. [ Conclusion] The study supplied theoretical basis for the application of phytase in livestock and poultry breeding.
