胰腺癌细胞对化疗药物敏感性的测定 被引量:9

Determination of sensitivity of pancreatic carcinoma cells to chemotherapy.

摘要 目的探讨人胰腺癌细胞株对常用化疗药物及其相互配伍的敏感性,为临床合理用药提供理论依据。方法将人胰腺癌细胞株(P_3、SW1990、Capan-2)细胞接种于96孔培养板,分别加入不同配伍及不同浓度的化疗药物,作用一定时间后采用噻唑蓝法(MTT法)测定其抗癌敏感性。结果以5-氟脲嘧啶和丝裂霉素为基本的三联用药在抑制率相当的情况下,其组成中各药物剂量仅为单一用药剂量的1/50;联合用药方案中以5-氟脲嘧啶、丝裂霉素、表阿霉素联合作用对胰腺癌细胞的抑制率最高,可达65.63%;随药物剂量增加和作用时间的延长,抑制率显著增高;化疗药物对不同胰腺癌细胞株细胞的抑制率存在显著的差异。结论联合用药化疗疗效明显优于单一用药,在可能条件下应增大药物剂量,延长药物作用时间,实行个体化用药方案,从而提高临床化疗效果。 Objective To investigate the sensitivity of human pancreatic carcinoma cell lines (P_3, SW1990, Ca-pan-2) to common chemotherapy drugs and to compatibility of medicines. Methods Human pancreatic carcinoma cells were inculated on a 96-well plate with addition of different compatibility of medicines and various concentrations of chemotherapy drugs. After a certain time for reaction, MTT method was used to detect the antitumor sensitivities. Results Under the circumstance of the same inhibiting rate, the single dose of the drugs in triple therapy (mainly including 5-FU and MMC) was only 1/50 of that of drugs used alone. In the regimens of the combined use of the chemotherapy drugs, the combined use of 5-FU, MMC and E-ADM revealed a highest inhibiting rate (65. 63%) to pancreatic carcinoma cells. With the increase of the drug dose and the prolongation of treatment, the inhibiting rate was obviously increased. There was a significant difference in inhibiting rate of the chemotherapy drugs against the various pancreatic carcinoma cells. Conclusion The therapeutic effects of combined use of the chemotherapy drugs was superior to that of single drug. The increase of the drug dose, prolongation of treatment and individuation of drug regimen might improve the clinical chemotherapy effects.
