肝纤维化和肝硬化患者肝内基质细胞衍生因子-1α水平的变化 被引量:7

A STUDY OF THE EXPRESSION OF SDF-1α IN INJURED HUMAN LIVER

摘要 目的观察肝纤维化和肝硬化患者肝内基质细胞衍生因子-1α(SDF-1α)表达情况。方法用ELISA法分别检测8例正常、10例肝纤维化、12例肝硬化病人肝组织内SDF-1α表达水平。结果正常对照组、肝纤维化组、肝硬化组的SDF-1α水平分别为(5.18±1.90)、(18.94±7.05)和(4.79±1.86)pg/mg。肝纤维化组的SDF-1α水平显著高于正常对照组和肝硬化组,而正常对照组和肝硬化组的SDF-1α水平无显著性差异。结论在肝纤维化阶段,肝脏仍具有较强的修复与再生能力,肝内表达SDF-1α增多,趋化肝干细胞向肝内定植并诱导其分化而修复肝损伤。肝硬化时,肝内SDF-1α表达水平低下,这或许是硬化肝再生与修复能力低下的重要因素,但SDF-1α水平低下的确切原因与意义还有待研究。 Objective To investigate the expression of stromal cell-derived factor-1α (SDF-1α) in different degree impaired liver, normal liver, fibrotic liver and cirrhosis. Methods Hepaiic tissue examples were collected from 30 cases : 8 patients with normal liver, 10 with fibrotic liver and 12 with cirrhosis. The SDF-1αexpression was assayed by ELISA method. Results The density of SDF-1α was (5.18 ± 1.90) pg/mg in normal liver, ( 18.94 ± 7.05 ) pg/mg in fibrotic liver and (4.79 ± 1.86 ) pg/mg in cirrhosis. The level of SDF-1α expression in the fibrotic liver was remarkably higher than that in the normal or sclerous liver, but there was no significant difference between that in the normal and in the sclerous liver. Conclusion In the earlier period of hepatic injury, the expression of SDF-1α in liver was up-regarded, which recruited the stem ceils to liver to participate in liver regeneration. But, it was low expression in the liver when the injured factors exist in long-term and the liver was damaged seriously. The exact mechanism of a low expression of SDF-1α in sclerous liver still need for further study.
