甘蓝型油菜小孢子胚状体诱导的主要影响因素研究 被引量:22

摘要 对基因型、小孢子发育时期、高温起始培养、培养基等甘蓝型油菜游离小孢子培养诱导胚状体的主要影响因素,研究表明:基因型和小孢子发育时期起决定性的作用,不同品种间甚至同一品种内的个体间,诱导频率差异显著;适宜培养的小孢子发育时期为单核晚期;培养基中加入活性炭可提高胚状体诱导频率;起始培养高温热击促进胚状体诱导。成熟胚状体在无激素B5培养基中,直接再生出植株或经次生胚继代培养再生植株。 The main factors of rape isolated microspore oulture such as genotype, microspore development stage, media, thermo shock, et al were studied. The results shows that genotype and the microspore development stage play a decisive role; cultural reaction and embroids among varieties even different plants in a same variety population differed significantly; late uninucleate stage when buds length reached 2.5~3.5mm was the most suitable one; active carbon in the media and 1~2 days thermo shock at the beginning culture were beneficial to the successful development of cultured microspores into embryos. The matured embryos derived from microspores cultured at B 5 media generated plants directly or through secondarily initiated embryos.
