摘要
以油菜小孢子培养得到的单倍体植株的离体组织作实验材料,诱导愈伤组织,获得生长良好的愈伤组织经不同剂量的60Co-γ射线辐射以后,部分分化成苗,对成苗单株进行AFLP分析,检测其诱变效应。结果表明茎段在附加有0.5mg/L2,4-D和2mg/L6-BA的MS培养基上能诱导出生长良好的愈伤组织;用3000~4000R辐射剂量对愈伤组织进行处理较为适宜;分子标记检测结果显示单倍体基因组在诱变过程中发生了不定向变异。
With the treatment of different doses of^(60)Co-γ irradiation on the callus in good state induced from the isolated tissue of the haploid Brassica napus plantlets, the irradiation effect was detected by the AFLP(Amplified Fragment-Length Polymorphism)markers on the plantlets from the irradiational callus. The results showed that the callus from the stem would grow well on the MS media with 0.5mg/L 2,4-D and 2mg/L 6-BA, the best dose of irradiation was ranged from 3 000 to 4 000 roentgen and there were undirectional variation occured in the callus during the γ-ray irradiation.
