摘要
为建立一种效率较高的芦笋花药培养诱导胚状体的机制,实验以TH IELIM F1为试材,利用正交实验设计L16(45)探讨以MS为基本培养基附加NAA、6-BA、2,4-D、蔗糖、谷氨酰胺等五种因素对芦笋花药培养诱导胚状体的影响。结果表明:NAA是影响芦笋花药培养中诱导胚状体的主要因素,其次是6-BA、蔗糖,而2,4-D、谷氨酰胺的影响较弱。芦笋花药培养诱导胚状体的最优组合是0.01 mg/L NAA、2.0 mg/L 6-BA、1.0 mg/L 2,4-D、50 g/L蔗糖、0 mg/L谷氨酰胺。
The aim of this study was to establish an efficient protocol for androgenetie embryoid induction in asparagus anther culture. Cuhivar THIELIM F, was used in the experiment with MS as the basal medium. Orthogonal arrays of L16 ( 4^5) were adopted to investigate the effect of NAA, 6 - BA, 2, 4 - D, sucrose, glutamine on response of embryoid induction. The results showed that NAA was the most effective element for embryoid induction in asparagus anther culture, followed by 6- BA and sucrose with the least effective for 2, 4 - D and glutamine. The optimized combination was derived with 0.01 mg/LNAA, 2.0 mg/L 6 - BA, 1.0 rag/ L 2, 4 - D, 5 g/L sucrose and 0 mg/L glutamine.
