HPV16 E7蛋白阳性和阴性乳癌组织中Rb和P16蛋白的表达 被引量:2

Expressions of Rb and P16 proteins in HPV16 E7 protein-positive and-negative breast carcinoma tissue

摘要 目的:研究人乳头瘤病毒16型(HPV16)E7蛋白阳性(E7+)和阴性(E7-)乳癌组织中Rb和P16蛋白的表达及意义。方法:以经PCR筛选证实的HPV16 DNA阳性的56例乳癌组织(经Western blotting证实42例HPV16E7+,14例E7-)为研究对象,采用Western blotting方法检测癌组织中Rb和P16蛋白的表达。结果:HPV16E7+、HPV16E7-乳癌组织中,Rb蛋白阳性率分别为9.5%(4/42)、50.0%(7/14),P16蛋白阳性率分别为85.7%(36/42)、21.4%(3/14),HPV16E+和E7-乳癌组织中P16和Rb蛋白阳性率差异均有统计学意义(χ2=20.525,10.898,P均<0.001)。HPV16E7+乳癌组织中,Rb和P16蛋白表达有关(rp=0.792,P=0.003);HPV16E7-乳癌组织中,Rb和P16蛋白表达无关(P=0.182)。结论:HPV16E7可通过降解Rb,促进P16蛋白表达,从而在乳癌发生发展中发挥作用。 Aim:To investigate the expression and significance of Rb and P16 proteins in human papillomavirus 16 type(HPV16) E7 protein-positive(E7+) and-negative(E7-) breast carcinoma tissues.Methods:A total of 56 HPV16 DNA-positive breast carcinoma specimens confirmed by PCR,including 42 HPV16 E7+ and 14 HPV16 E7-breast carcinoma specimens certified by Western blotting assay,were subjected to detect Rb and P16 proteins by Western blotting assay.Results:The positive rates of Rb in HPV16 E7+ and HPV16 E7-breast carcinoma tissues were 9. 5% (4/42) and 50.0% (7/14), respectively, while the positive rates of P16 were 85.7% (36/42) and 21.4% (3/14), respectively. There were significant differences in the expressions of P16 and Rb between HPVI6 E7^+ and HPV16 E7 - tissue (χ2 was 20. 525,10. 898, respectively, P 〈 0.01 ). The expression of P16 was also associated with that of Rb in HPV16 E7^+ tissue (rp =0.792,P =0.003) ,but not in HPV16 E7- breast carcinoma tissue (P=0. 182). Conclusion: The HPV16 E7 may play an important role in the carcinogenesis and progression of breast carcinoma by degrading Rb protein and up-regulating P16 protein expression.
