摘要
测定了小牛软骨血管生成抑制因子(cartilageangiogenesisinhibitoryfactor,CAIF)对血管内皮细胞的细胞毒作用、对内皮细胞骨架系统及其运动迁移的抑制效应、对小鼠肿瘤生长的抑制效应.结果表明CAIF对血管内皮细胞产生明显的细胞毒作用,对内皮细胞骨架具有明显的抑制作用,能显著抑制内皮细胞的运动迁移,并显著抑制小鼠肿瘤的生长.提示CAIF的抗肿瘤作用机制可能是:通过抑制血管内皮细胞的骨架系统及其分裂增殖,并抑制内皮细胞的运动迁移,从而抑制血管内皮芽的形成和新生血管的生成.由于切断了肿瘤的营养供应及血道转移途径,从而抑制肿瘤的生长和转移.
It was previously shown that cartilage angiogenesis inhibitory factor (CAIF) has an inhibitory effect on angiogenesis. The inhibitory effect of CAIF on the growth of bovine artery endothelial cells was studied using cytotoxity assay. The effect of CAIF on the skeleton of endothelial cells and Hela cells was investigated by whole cell scanning electron microscopy. The effect of CAIF on endothelial cell migration and the tumor growth were quantified by ordinary methods. The results showed that CAIF exerted inhibitory effects on the growth and migration of endothelial cells, and made the skeleton of endothelial cells wrinkled, but had no significant effect on that of Hela cells. CAIF also inhibited the tumor growth implanted on mice. These results suggest that CAIF is a specific angiogenesis inhibitory factor, and it inhibits the DNA synthesis and the skeleton of endothelial cells, so inhibits their growth and migration, then inhibits angiogenesis, 'Tumor growth is angiogenesis dependent', CAIF inhibit. tumor growth significantly.
