糖基转移酶基因β3GalT7/GnT8和β3GnT2在白血病细胞株及人骨髓有核细胞中的表达 被引量:1

摘要 目的:从mRNA水平观察不同白血病细胞和人骨髓标本中糖基转移酶β3GalT7/GnT8和β3GnT2的表达情况。方法:采用RT-PCR技术,在8种白血病细胞株及24例白血病患者骨髓有核细胞中检测β3GalT7/GnT8,β3GnT2 mRNA水平的表达,以10例正常成人骨髓有核细胞作相对正常对照。结果:β3GalT7/GnT8在SHI-1,THP-1,HL-60,K562,NB4,U937细胞和18例初诊白血病患者骨髓标本中有不同程度的表达,在KG1,Raji细胞和相对正常对照中没有表达;β3GnT2在SHI-1,THP-1,HL-60,K562,NB4,U937,Raji细胞,4例相对正常对照和16例初诊白血病患者骨髓标本中有不同程度的表达,在KG1细胞中和6例相对正常对照中没有表达。结论:β3GalT7/GnT8和β3GnT2与白血病的发生发展存在一定的关系,其mRNA的表达水平可以作为白血病早期诊断的一个标志。 Objective: To observe the expression of β3GalT7/GnT8 and β3GnT2 gene in different leukemia cells and patients on mRNA level. Methods: RT-PCR was used to detect the mRNA expression in 8 leukemia cells ,24 patients and bone marrow of 10 normal persons. Results: Different expression level of 3GalT7/GnT8 can be found in leukemia cells SHI-1, THP-1, HL-60, K562, NB4, U937 and 18 patients who were leukemia at first diagnosis, but never found in leukemia cell Raji, KG1 and bone marrow of normal persons ; β3GnT2 expressed in SHI-1, THP-1, HL-60, K562, NB4, U937, Raji, 16 patients and 4 bone marrow of normal persons, never found in KG1 and 6 bone marrow of normal persons. Conclusion: β3GalT7/GnT8 and β3GnT2 expression level were related to diagnosis of leukemia.
