摘要
目的:观察针刺"肺俞"、"灵台"穴对新型抗癌药物紫杉醇在lew is肺癌小鼠体内分布趋势的影响,为化疗药物靶向给药的研究提供新思路,进一步验证"针药并举"协同增效的机理。方法:选用C57BL/6小鼠建立lew is肺癌模型,选用单纯腹腔注射紫杉醇以及腹腔注射紫杉醇分别与针刺"肺俞"、"灵台"穴位相结合等3种方法进行治疗,在接种Lew is肺癌瘤株后第13天,分别于注射药物后1、2、3h将各实验组动物处死,取肺、脾、肝、肾组织,用高效液相法分别检测在不同时间点不同组织中的紫杉醇含量。结果:(1)各实验组不同组织中紫杉醇的代谢趋势:药物组肺、肝、肾组织中的药物含量在2h时达高峰,之后明显下降;脾组织药物含量在不同时间点呈下降趋势。针药肺俞组和针药灵台组肺、肾中药物含量在2h时达高峰;肝脏中药物含量在3个时间点呈升高趋势;脾中药物含量逐渐下降。(2)不同组织中药物含量比较:药物组肾、脾两脏中紫杉醇含量较高,其次为肝脏、肺脏;两针药组并未表现出这种规律,在不同时间点肺中药物含量甚至分别高于肝、脾、肾三脏。(3)不同时间点组织中药物含量比较:针药肺俞组肺组织中药物含量在3个时间点均明显高于药物组;针药灵台组肺组织中紫杉醇含量在2h时高于药物组;同时,除3h时针药肺俞组肾组织中药物含量与药物组无显著差异外,两针药组脾、肾中的紫杉醇含量在其它各时间点均明显低于药物组。结论:(1)针刺"肺俞"、"灵台"穴影响了紫杉醇在小鼠体内的代谢,使药物在各脏器的分布发生明显变化。(2)针刺"肺俞"、"灵台"使药物在肝脏达高峰的时间延长,表明针刺延长了药物在体内的停留时间,即延长了药物在体内的作用时间。(3)针刺"肺俞"、"灵台"穴能使紫杉醇在肺脏的含量增加,在脾、肾两脏的含量减少,说明针刺对紫杉醇在Lew is肺癌小鼠体内的分布具有趋向性影响,产生靶向效应。(4)针刺"肺俞"穴使药物在肺脏的含量连续高于药物组,说明"肺俞"穴对药物在肺脏的趋向性要强于"灵台"穴。
Objective:The experiment establish Lewis lung cancer mouse model and survey the effects of Feishu and Lingtai aeupoint acupuncture on anticancer drug paclitaxel's body distribution tendency, to provide new ideas for target administration of Chemotherapeuties, and to authenticate furtherly synergism mechanism of developing acupuncture and drug simultaneously. Methods : Lewis lung cancer mouse model was established by using C57 BL/6 mouse. The therapy methods included intraperitoneal injection paclitaxel merely and combination intraperitoneal injection paclitaxel with Feishu and Lingtai aeupoint acupuncture respectively. On the 13th day after innoculation of Lewis lung cancer strain, sacrifice of animal in experiment groups at 1 h, 2h, 3h respectively after drug injection, lung, spleen, hver and kidney tissues were obtained and drug contents in different tissues were detected respectively by HPLC. Drug concentrations of samples were calculated by normalization. Results: ( 1 ) The metabolism tendency of paclitaxel in different tissues of every experimental group:In drug group, drug contents of lung, liver and kidney tissues reached the peak at 2h, then decreased obviously; drug contents of spleen tissue showed downtrend at different time points and the lowest content at 3h. In acupuncture Feishu or Lingtai acupoint + drug groups, drug contents of lung, kidney tissues reached the peak at 2h; drug contents of liver had a gradual increase tendency at three time points and the highest content at 3h ; drug contents of spleen fail-off gradually. (2) The comparison of drug contents in different tissues : in drug group, paclitaxel contents of kidney, spleen were higher, next was liver, the least was lung. In two acupuncture + drug groups, there was no show of this kind of rule. At different time points, drug contents of lung was even higher than that of liver,spleen and kidney, respectively. (3) The comparison of drug contents in tissues at different time points : Drug contents of lung in acupuncture Feishu acupoint + drug group was obviousely higher than that of drug group at three time points. Paclitaxel contents of lung in acupuncture Lingtai acupoint + drug groups were significantly higher than that in drug group at 2h. Simultaneously,paclitaxel contents of spleen,kidney in two acupuncture + drug groups were significantly lower than that in drug group, except that there were no significant deviation of drug contents in kidney between acupuncture Feishu acupoint + drug group and drug group alone at 3h. Conclusion : The research findings of experiment show the following points : ( 1 ) acupuncture of Feishu or Lingtai acupoint influenced the metabolism of paclitaxel in mouse and make drug disposition in each organ marked change. (2) Acupuncture of Feishu or Lingtai acupoint can extend the time of drug reaching a peak in liver. It indicates that acupuncture may prolong detention time of drug in vivo, which means it lengthen action time of drug in vivo. (3) Acupuncture of Feishu or Lingtai acupoint can increase the paclitaxel contents in lung and decrease the contents in spleen and kidney. It illustrates that acupuncture may make tropism effect on the disposition of paclitaxel in Lewis lung cancer mouse and produce the target effect. (4) Drug contents of lung in acupuncture Feishu acupoint + drug group was higher than that of drug group at three time points. That means :The ability of target distribution to lung of Feishu point is stronger than Lingtai point.
