摘要
目的观察并比较电针"大肠俞""天枢"穴对腹泻型肠易激综合征(D-IBS)模型大鼠内脏高敏反应、炎症因子及脑肠肽的影响,探讨不同经脉和不同穴性穴位的效应特异性。方法将40只SPF级雄性Wistar大鼠随机分为空白组,模型组,大肠俞组,天枢组,每组10只,急性应激与慢性应激相结合CAS方法制备D-IBS大鼠模型。空白组不予以任何操作,模型组只造模不针刺,大肠俞组和天枢组从造模一周后分别给予双侧"大肠俞"和"天枢"穴电针干预,连续28天。检测大鼠内脏高敏反应、采用酶联免疫吸附法(ELISA)测定大鼠血清中的IL-1β、IL-10、TNF-α、5-HT、SP、CGRP的含量。结果与空白组相比,模型大鼠的AWR评升高(P<0.05),血清IL-1β、TNF-α、5-HT、SP、CGRP的水平显著升高(P<0.05),IL-10含量显著下降(P<0.05);与模型组相比,大肠俞组及天枢组大鼠的AWR评分有降低趋势(P<0.05),血清IL-1β、TNF-α、5-HT、SP、CGRP水平显著下降(P<0.05),IL-10含量显著升高(P<0.05);与大肠俞组相比,天枢组在的AWR评分显著降低(P<0.05),血清IL-1β、TNF-α、5-HT、SP、CGRP水平显著下降(P<0.05),IL-10含量显著升高(P<0.05)。结论电针"大肠俞""天枢"穴均可改善IBS-D大鼠的症状,但电针"天枢"穴的调节作用较"大肠俞"穴更明显。
Objective To observe and compare the effects of electroacupuncture at Dachangshu(BL25)and Tianshu(ST25)on visceral hypersensitivity,inflammatory factors and brain intestinal peptides in model rats with diarrhea-type irritable bowel syndrome(D-IBS)and explore the effect specificity of different meridians and acupoints.Methods Forty male SD rats were randomly divided into blank group,model group,Dachangshu(BL25)group and Tianshu(ST25)group,with 10 rats in each group.D-IBS rat model was prepared by CAS method combining acute stress with chronic stress.Blank group was given no operation.The model group was not given acupuncture.Electroacupuncture at Dachangshu(BL25)and Tianshu(ST25)were applied in the corresponding groups 28 times after a week of modeling.The abdominal drawal reflex(AWR)scores were measured.The levels of IL-1β,IL-10,TNF-α,5-HT,SP and CGRP were detected by modified ELISA.Results Compared with those of the normal group,AWR scores,levels of IL-1β,TNF-α,5-HT,SP and CGRP in serum were increased significantly(P<0.05)and IL-10 in serum of the model group was decreased significantly(P<0.05).Compared with those of the model group,AWR scores,levels of IL-1β,TNF-α,5-HT,SP and CGRP in serum of the Dachangshu(BL25)group and the Tianshu(ST25)group were decreased significantly(P<0.05),while IL-10 in serum of the Dachangshu(BL25)group and the Tianshu(ST25)group were increased significantly(P<0.05).Compared with the Dachangshu group,AWR scores,levels of IL-1β,TNF-α,5-HT,SP and CGRP in serum of the Tianshu(ST25)group were decreased significantly(P<0.05),while IL-10 in serum of the Tianshu(ST25)group was increased significantly(P<0.05).Conclusion Electroacupuncture at Dachangshu(BL25)or Tianshu(ST25)can improve the symptoms of D-IBS rats.The effect of electroacupuncture at Tianshu(ST25)is better.
