摘要
目的:探讨实验性肝细胞增生酶异常灶量化指标的检测方法。方法:应用抑制物抑制二乙基亚硝胺(NDEA)致大鼠肝细胞增生酶异常灶(γ-谷氨酰转肽酶阳性灶和胎盘型谷胱甘肽s-转移酶阳性灶)发生的短期实验模型以及电脑化图形扫描分析系统对肝细胞增生酶异常灶进行测量,统计比较其结果。结果:三个肝叶切片的总值和任一肝叶切片数值以及各肝叶切片数值之间比较差异不显著;评价抑制物抑制γ-谷氨酰转肽酶(γ-GT)灶发生情况时,以右前叶数值更接近三叶总值;不同人员处理相同的蜡块,所得结果在单位面积的灶数(个/mm2)、灶面积密度(mm2/cm2)上相比差异不显著;γ-GT与胎盘型谷胱甘肽S-转移酶(GSTP)染色结果比较差异显著,GSTP数值大于γ-GT。结论:评价肝细胞增生酶异常灶的发生时,以右前叶代表性最强;我们的观察方法和所得的结果重复性好;GSTP染色较γ-GT染色更准确并可节省工作量,是一种较优越的染色方法。提示研究大鼠肝细胞增生酶异常灶时,选择右前叶作为检测对象,GSTP作为阳性标志酶更简便可行。
Objective: To investigate the quantitative analysis method of experimental enzyme altered hyperplastic hepatocyte foci. Method: To study the Diethylnitrosamineinduced enzyme altered hyperplastic hepatocyte foci positive for γglutamyltranspeptidase (γGT) and Glutathione Stransferase placental form (GSTP) in male Sprague Dawley rats by means of the short term in vivo test model, measuring and calculating them with computerized scanning image analysis system and to compare their results. Results: There was no significant difference among the values from three liver lobes and that from any one lobe. Evaluating the inhibitory effect of inhibitor on the γGT positive hepatocyte foci, it was found that the values of the right anterior lobe were closest to that of three lobes. Different researchers dealed with the same wax blocks with our experimental method, the results were stable. The values of GSTP stain were significantly larger than that of γGT stain. Conclusion: Our experimental technique and quantitative analysis method were stable and repeatable. The occurrence of enzyme altered foci could mostly be represented by the right anterior lobe. The expression of GSTP was stronger than that of γGT. It is convenient for us to select the right anterior lobe as the representative sample and GSTP is a good positive marker for studying chemical carcinogeninduced hepatocarcinogenesis in rats.
