摘要
目的:提取并纯化豚鼠脑、脊髓髓鞘碱性蛋白(MBP),对其理化特征和致实验性自身免疫性脑脊髓炎(EAE)的活性进行鉴定,并初步探讨口服MBP诱导免疫耐受防治EAE的作用。方法:以豚鼠脑、脊髓为原料,通过去脂、抽洗、酸溶解、高速离心等步骤提取MBP,并在Wistar大鼠中诱导EAE和口服耐受。结果:所提取的MBP具有与文献报告结果类似的理化特征和诱发EAE的活性;通过口服MBP,成功诱导出对EAE的免疫耐受。结论:MBP对研究EAE的发病机制和病理变化及通过口服MBP诱导免疫耐受对EAE和多发性硬化症的防治具有重要意义。
ABSTRACT Objective: To study physical and chemical characteristics and biological activity of myelin basic protein(MBP) from guinea pig brain and spinal cord. To investigate the activity of MBP in inducing experimental autoimmune encephalomyelitis(EAE) and suppression of EAE by oral administration of MBP. Methods: By means of defatting with methanol-chloroform, acid extraction and centrifugation , MBP was obtained . MBP was emulsified in complete Freund's adjuvant(CFA)and was used to induce EAE and oral tolerance in Wistar rats. Results: EAE was successfully induced with MBP plus adjuvant in Wistar rats. In addition, MBP was found to have protective action and therapeutic effect on EAE when animals were fed MBP before or after disease induction. Conclusion: MBP can be used to induce immunologic tolerance in the prevention and treatment of EAE.
