摘要
目的评价阿司匹林对SD大鼠的母体及胎仔毒性。方法将32只SD孕鼠随机分为2个组(每组16只):阿司匹林组(280mg/kg)及对照组,阿司匹林组大鼠于受孕的第7~16天灌胃给予阿斯匹林(溶剂为1.5%羧甲基纤维素钠),对照组给予1.5%羧甲基纤维素钠,灌胃量均为每天1.0mL/100g.BW。受孕的0、7、12、16、20d称量体重,在妊娠的第20天,颈椎脱臼处死,计数活胎数、死胎数和吸收胎数;观察胎仔外观、称量其体重后,将每窝1/2的活胎仔乙醇固定、氢氧化钾消化、茜素红染色、甘油透明后检查骨骼发育情况。另1/2活胎仔经Bouin’s液固定后,检查内脏发育情况。结果受试后阿司匹林组SD大鼠体重下降,16d和20d孕鼠体重及终期体重增重与对照组比较,差异均有统计学意义(P<0.01);阿司匹林组SD大鼠平均活胎数减少,吸收胎数及死胎数增加,活胎平均体重降低,身长和尾长缩短,各指标与对照组比较,差异均有统计学意义(P<0.01);外观畸形率、骨骼和内脏畸形率明显增加,与对照组比较,差异有统计学意义(P<0.05)。结论该实验条件下阿司匹林(280mg/kg)对SD大鼠可能有明显的母体毒性和胚胎的致畸毒性。
Objective To study the embryo toxicity and teratogenecity of acetylsalicylic acid in SD rats. Methods The pregnant SD rats were divided into two groups, acetylsalicylic acid group (280 mg/kg) and the control group. Every group consisted of 16 pregnant rats. The rats of both groups were administrated with acetylsalicylic acid and 1.5% sodium carboxymethylecellulose ( CMC - Na) 1.0 mL/100g · BW orally during the period of organ formation (from day 7 to day 16), once a day. The pregnant rats were weighted in day 0, 7, 12, 16 and 20, and were killed by cervical vertebral dislocation in day 20, and then the numbers of the living embryos, dead fetus and absorbed fetus were counted. The appearances of the embryos were examined, and tail lengths and weights were measured. After that, half of living embryos were used to study the bone development after ethanol fixing, potassium hydroxide melting, Alizarin Red staining and glycerin transparencing. The other living embryos were examined for the visceral development after Bouin's solution fixing. Results Compared with the control group, the weights of the pregnant rats decreased significantly in the acetylsalicylic acid group in day 16 and day 20 (P 〈 0. 01 ). In the acetylsalicylic acid group, the number of the living embryos decreased, while the resorbed fetus and the dead fetus increased significantly. The living embryos'weight, body length and tail length decreased significantly com- pared to these in the control group ( P 〈 0. 01 ). The rate of the appearance abnormalities, the deformity rate of the skeleton and internal organs increased significantly in the acetylsalicylic acid group ( P 〈 0. 05 ). Conclusion Acetylsalicylic acid (280 mg/kg) had embryonic and teratogenic toxicity in SD rats.
